چکیده

Exogenous purines are known to suppress purine synthesis de novo in bacteria (2-4), HeLa and L cells (5), and ascites cells (6) growing in culture. High levels of purines, especially adenine, inhibit accumulation of late purine precursors in the media of bacteria with blocks in the pathway of purine synthesis (2, 3), or of formylglycinamide ribonucleotide in Ehrlich ascites cells grown in the presence of aaaserine (6). These studies have implicated an early step in purine biosynthesis as the site of a feedback regulatory mechanism. Studies from this laboratory have shown that this inhibition operates on the first specific reaction of purine biosynthesis in which ribosylpyrophosphate 5-phosphate and glutamine interact to form phosphoribosylamine, and that adenosine triphosphate is the most potent of the inhibitory purine ribonucleotides (7, 8). Several purine analogues have also been shown to affect purine synthesis de novo. Gots and Gollub (9) observed that 6-mercaptopurine and 6-thioguanine inhibited accumulation of 5amino-4-imidazolecarbosamide ribonucleoside by Escherichia coli B-96 grown on limiting concentrations of adenine. Sartorelli and Le Page (10) and Le Page and Jones (1 I) observed that purinethiols inhibited accumulation of formylglycinamide ribonucleotide in Ehrlich ascites cells or mouse neoplasms treated with azaserine. Simpson, Bennett, and Golden (12) described the inhibition of incorporation of formate-‘4C into purines in Ehrlich ascites cells under conditions in which the terminal steps of purine synthesis were not inhibited. All of these studies suggested that these purine analogues might also be inhibiting purine synthesis by acting on an early reaction of the sequence. This paper presents evidence that the ribonucleotide derivatives of 6-mercaptopurine, 6-thioguanine, %azaguanine, and 4-hydrosypyrazolo-(3,4-d)pyrimidine effect a pseudofeedback inhibition of glutamine ribosylpyrophosphate 5-phosphate amidot.ransferase, mimicking the inhibit.ory action of natural purine ribonucleotides on this enzyme.

Purines, Purine Nucleosides, and Analogues

Of 142 purines, purine nucleosides, and analogues tested for inhibition of growth of Escherichia coli B Hill, 45 were active. Of these, 27 were evaluated for inhibition of other E. coli lines, including those resistant to 6-thioguanine, 2-fluoroadenosine, 2,6-diaminopurine, or 6-mercaptopurine. Most toxic to the parent lines were 2-fluoroadenosine, 2-fluoroadenine, 2-fluoro-5'-deoxyadenosine, a...

Resistance to purine antagonists in experimental leukemia systems.

Resistance to purine analogs by loss of nucleotide-forming capacity was observed in a number of experimental tumor systems. Such resistance frequently was accom panied by loss of specific purine ribonucleotide pyrophosphorylase activities. How ever, it is evident that other mechanisms of resistance to purine analogs also exist both in mouse leukemia and in human leukemia. Attention was focused ...

Identification of the antineoplastic agent 6-mercaptopurine as an activator of the orphan nuclear hormone receptor Nurr1.

The purine anti-metabolite 6-mercaptopurine is one of the most widely used drugs for the treatment of acute childhood leukemia and chronic myelocytic leukemia. Developed in the 1950s, the drug is also being used as a treatment for inflammatory diseases such as Crohn's disease. The antiproliferative mechanism of action of this drug and other purine anti-metabolites has been demonstrated to be th...

Inhibition of de novo purine biosynthesis and interconversion by 6-methylpurine in Escherichia coli.

The inhibition of Escherichia coli strain B and strain W-11 by 6-methylpurine depended on the formation of 6-methylpurine ribonucleotide by the action of adenine phosphoribosyltransferase (AMP: pyrophosphate phosphoribosyltransferase, EC 2.4.2.7). 6-Methylpurine ribonucleotide inhibited the de novo synthesis of purines, presumably via pseudofeedback inhibition of phosphoribosylpyrophosphate ami...

Mechanisms of action of 6-thioguanine, 6-mercaptopurine, and 8-azaguanine.

The effects of 6-thioguanine on purine biosynthesis and cell viability have been examined in H.Ep. 2 cells grown in culture. Toxicity is not reversed by aminoimidazolecarboxamide, suggesting that inhibition of purine biosynthesis de novo is not the sole mechanism of toxicity. Also, 6-(methylmercapto)purine ribonucleoside, a potent inhibitor of purine biosynthesis de novo, produces more marked r...
